چکیده

Transgenic animals, mice and rats, harboring temperature-sensitive simian virus 40 large T-antigen have been found to be very useful for establishing immortalized cell lines from tissues that have been proved difficult to culture in vitro. Here, we succeeded in establishing a conditionally immortalized tracheal epithelial cell line RTEC11 from adult transgenic rats harboring the oncogene. Morphological and functional investigations demonstrated that the RTEC11 cells were polarized epithelial cells maintaining a regulated permeability barrier function. Although the large T-antigen was expressed in the nuclei and the cells grew continuously at a permissive temperature of 33 ̊C, the down regulation of large T-antigen at a nonpermissive temperature of 39 ̊C was associated with cell growth arrest and cellular differentiation. In addition, global gene expression and computational network analyses indicated that a significant genetic network including CDKN1A was associated with cellular differentiation induced by inactivation of the large T-antigen. The tracheal epithelial cell line RTEC11 with unique characteristics should be useful as an in vitro model of the tracheal epithelium for physiological, pharmacological and toxicological investigations. Moreover, experiments using an immortal cell line may offer an alternative to experiments using living animals.

Development and characterization of conditionally immortalized gastric epithelial cell lines from transgenic rats harboring temperature-sensitive simian virus 40 large T-antigen gene.

Conditionally immortalized gastric epithelial cell lines were established from transgenic rats harboring temperature-sensitive simian virus 40 (tsSV40) large T-antigen gene. Gastric mucosal cells and epithelial tissues isolated from the stomach of the transgenic rats were cultured at permissive temperature (33 degrees C), and proliferative cells were cloned by colony formation. Six cell lines (...

Development of Oral Epithelial Cell Line ROE2 with Differentiation Potential from Transgenic Rats Harboring Temperature-Sensitive Simian Virus40 Large T-Antigen Gene

We have developed an immortalized oral epithelial cell line, ROE2, from fetal transgenic rats harboring temperature-sensitive simian virus 40 large T-antigen gene. The cells grew continuously at either a permissive temperature of 33°C or an intermediate temperature of 37°C. At the nonpermissive temperature of 39°C, on the other hand, growth decreased significantly, and the Sub-G1 phase of the c...

Morphological and functional characterization of a conditionally immortalized collecting tubule cell line.

A conditionally immortalized collecting tubule cell line, mCT1, was derived from the H-2Kb-ts A58 transgenic mouse (ImmortoMouse), which harbors a temperature-sensitive mutant of the SV40 large T antigen oncogene. Cells maintained under permissive conditions [33°C with interferon-γ (IFN-γ)] form epithelial monolayers, express large T antigen, and proliferate (>50 passages). The cells retain pro...

Characterization of Murine Thymic Stromal-Cell Lines Immortalized by Temperature-Sensitive Simian Virus 40 Large T or Adenovirus 5 E1a

The heterogeneity of thymic stromal cells is probably related to their role in providing different microenvironments where T cells can develop. We have immortalized thymic stromal elements using recombinant retroviral constructs containing a temperature-sensitive simian virus 40 (SV40tsA58) large-T antigen gene or the adenovirus 5 E1a region linked to the gene coding for resistance to G418. Cel...

Direct derivation of conditionally immortal cell lines from an H-2Kb-tsA58 transgenic mouse.

Studies on cell lines have greatly improved our understanding of many important biological questions. Generation of cell lines is facilitated by the introduction of immortalizing oncogenes into cell types of interest. One gene known to immortalize many different cell types in vitro encodes the simian virus 40 (SV40) large tumor (T) antigen (TAg). To circumvent the need for gene insertion in vit...
